Abstract

Lobster bambu (Panulirus versicolor) merupakan komoditas krustasea bernilai ekonomi tinggi yang berpotensi menjadi pembawa berbagai agen penyakit, termasuk White Spot Syndrome Virus (WSSV). Penelitian ini bertujuan mendeteksi keberadaan WSSV dan menentukan prevalensinya pada lobster bambu yang dilalulintaskan melalui sistem karantina di Jawa Timur. Penelitian menggunakan metode deskriptif dengan pendekatan molekuler terhadap 20 ekor lobster bambu asal Sumenep, Madura, yang diperiksa pada periode November–Desember 2025. Jaringan insang dan pleopod diekstraksi menggunakan silica extraction kit dan dianalisis menggunakan nested Polymerase Chain Reaction (PCR) dengan primer 146F1/146R1 dan 146F2/146R2. Produk amplifikasi divisualisasikan melalui elektroforesis gel agarosa 1,5%. Hasil pengamatan klinis menunjukkan seluruh lobster tampak sehat tanpa gejala khas infeksi WSSV. Namun, hasil nested PCR mendeteksi satu sampel positif yang ditandai oleh pita DNA berukuran sekitar 941 bp, sedangkan 19 sampel lainnya menunjukkan hasil negatif. Berdasarkan hasil tersebut, prevalensi WSSV pada lobster bambu sebesar 5%. Temuan ini menunjukkan bahwa lobster bambu yang tampak sehat berpotensi berperan sebagai pembawa subklinis WSSV. Oleh karena itu, penerapan skrining molekuler secara rutin diperlukan untuk mendukung biosekuriti dan pengawasan lalu lintas komoditas krustasea melalui sistem karantina.
